About FOXO4-DRI
FOXO4-DRI is a synthetic D-retro-inverso peptide: built from mirror-image D-amino acids in reversed sequence, making it highly resistant to enzymatic breakdown. It competes with endogenous FOXO4 for binding p53, and in senescent cells that competition releases p53 to trigger the apoptosis those cells normally resist.
The 2017 Cell publication on FOXO4-DRI is foundational senolytic literature: naturally aged mice showed restored running behaviour, coat density and renal function after senescent-cell clearance. Senolytics have since become one of the most active areas of aging research.
